After making their way through high school (twice), big changes are in store for officers Schmidt (Jonah Hill) and Jenko (Channing Tatum) when they go deep undercover at a local college. But when Jenko meets a kindred spirit on the football team, and Schmidt infiltrates the bohemian art major scene, they begin to question their partnership. Now they don't have to just crack the case - they have to figure out if they can have a mature relationship. If these two overgrown adolescents can grow from freshmen into real men, college might be the best thing that ever happened to them.
22 Jump Street, film diretto da Phil Lord e Chris Miller, continua a seguire le avventure dei poliziotti Schmidt (Jonah Hill) e Jenko (Channing Tatum). Dopo il successo della missione precedente, i due agenti vengono riassegnati al programma sotto copertura gestito dal Capitano Dickson (Ice Cube), ora situato al numero 22 di Jump Street. Schmidt e Jenko vengono quindi spediti in un college, dove - fingendosi studenti - devono scoprire chi è lo spacciatore che vende una droga denominata WhyPhy, che ha causato la morte di una ragazza.
Sotto copertura, i poliziotti iniziano a integrarsi con i giovani nel campus: l'atletico Jenko fa amicizia con gli sportivi Zook e Rooster, con cui si reca ai party delle fraternite. L'impacciato Schmidt invece conosce la studentessa d’arte Maya (Amber Stevens West), per cui inizia a provare una certa attrazione. La vita spensierata del college prende il sopravvento e man mano che le indagini proseguono, portando pochi risultati, l’amicizia tra i due agenti inizia a deteriorarsi
