Il gruppo di amici formato da Phil (Bradley Cooper), Stu (Ed Helms), Alan (Zach Galifianakis) e Doug (Justin Bartha) parte per recarsi in Thailandia, dove si celebrerà il matrimonio di Stu. Visto come è andata con l'addio al celibato a Las Vegas ( narrato nel precedente film), Stu si cautela e decide che i festeggiamenti culmineranno con un tranquillo brunch domenicale, rigorosamente separato dalla cerimonia. Ma ogni calcolo viene disatteso da un mix di eventi potentissimo.

Sono passati due anni dopo quello che è accaduto prima a Las Vegas e poi in Thailandia. Phil (Bradley Cooper), Stu (Ed Helms) e Doug (Justin Bartha) vivono tranquilli e senza particolari eventi che scuotano le loro esistenze mentre Alan (Zach Galifianakis) è l'unico del gruppo di vecchi amici che non ha ancora trovato uno scopo nella vita. Quando una crisi personale colpisce Alan e lo spinge a chiedere l'aiuto di cui ha bisogno, Phil, Stu e Doug non mancheranno di accompagnarlo in un viaggio on the road, durante il quale ne vedranno delle belle.
